Section 14.5 - Fluorescent and Biotinylated Dextrans

چکیده

Dextrans are hydrophilic polysaccharides characterized by their moderate to high molecular weight, good water solubility and low toxicity. They are widely used as both anterograde and retrograde tracers in neurons 1,2 and for numerous other applications. Dextrans are biologically inert due to their uncommon poly-(α-D-1,6-glucose) linkages, which render them resistant to cleavage by most endogenous cellular glycosidases. They also usually have low immunogenicity. Molecular Probes offers almost 100 fluorescent and biotinylated dextran conjugates in several molecular weight ranges.

Multiple neuroanatomical tracing in primates.

The present report deals with a multiple tract-tracing procedure in non-human primates enabling the simultaneous visualization of retrogradely transported Fluoro-Gold (FG) and cholera toxin B subunit (CTB) in combination with anterogradely transported biotinylated dextran amine (BDA). Two issues have played key roles on the achievement of this reliable procedure: first, the recent development o...

Resolving the Detailed Structure of Cortical and Thalamic Neurons in the Adult Rat Brain with Refined Biotinylated Dextran Amine Labeling

Biotinylated dextran amine (BDA) has been used frequently for both anterograde and retrograde pathway tracing in the central nervous system. Typically, BDA labels axons and cell somas in sufficient detail to identify their topographical location accurately. However, BDA labeling often has proved to be inadequate to resolve the fine structural details of axon arbors or the dendrites of neurons a...

The sensorimotor slice.

The reciprocal connections between primary motor cortex (M1) and primary somatosensory cortex (S1) are hypothesized to play a role in an animal's ability to update its motor plan in response to changes in the sensory periphery. These interactions provide the sensory cortex with anticipatory knowledge of motor plans. In the mouse neocortex there are representations of the body surface within bot...

Rostral ventromedial medulla connections in Cavia porcellus and their relation with tonic immobility defensive behavior: A biotinylated dextran amine neurotracing study

The rostroventromedial medulla (RVM), together with the periaqueductal gray matter (PAG), constitutes the descendent antinociceptive system. Additionally, these structures mutually regulate defensive behaviors, including tonic immobility (TI) in guinea pigs. The current study was undertaken to evaluate the connections of the RVM with the PAG and the spinal cord in guinea pigs in order to provid...

Transsynaptic Tracing from Peripheral Targets with Pseudorabies Virus Followed by Cholera Toxin and Biotinylated Dextran Amines Double Labeling.

Transsynaptic tracing has become a powerful tool used to analyze central efferents that regulate peripheral targets through multi-synaptic circuits. This approach has been most extensively used in the brain by utilizing the swine pathogen pseudorabies virus (PRV)(1). PRV does not infect great apes, including humans, so it is most commonly used in studies on small mammals, especially rodents. Th...
